Microglial heterogeneity in chronic pain

چکیده

In this review, we report existing preclinical evidence on how the CNS compartment as well sex affect microglia functions in health. We highlight that recent advances transcriptomics analyses have led to thorough characterization of disease-associated microglial states mice and humans. then consider specific scenario peripheral nerve or tissue injury which induce expression a subset genes dorsal horn spinal cord. suggest intriguing possibility future studies may disclose existence unique transcriptional profile is associated with chronic pain conditions. also collect activation pain-related areas brain can be observed models neuropathic agreement neuroimaging patients. Based discussed here, predict neuroimmune interactions should complement our current understanding functions, but adventure using novel approaches such scRNA-seq, spatial transcriptomics, CYTOF transmission electron microscopy provide more complete function, transcriptome structure pain.

Persistent Alterations in Microglial Enhancers in a Model of Chronic Pain.

Chronic pain is a common and devastating condition that induces well-characterized changes in neurons and microglia. One major unanswered question is why these changes should persist long after the precipitating injury has healed. Here, we suggest that some of the longer-lasting consequences of nerve injury may be hidden in the epigenome. Cell sorting and sequencing techniques were used to char...

p38 MAPK, microglial signaling, and neuropathic pain

Accumulating evidence over last several years indicates an important role of microglial cells in the pathogenesis of neuropathic pain. Signal transduction in microglia under chronic pain states has begun to be revealed. We will review the evidence that p38 MAPK is activated in spinal microglia after nerve injury and contributes importantly to neuropathic pain development and maintenance. We wil...

Microglial cathepsin B contributes to the initiation of peripheral inflammation-induced chronic pain.

Interleukin (IL)-1β and IL-18 play critical roles in the induction of chronic pain hypersensitivity. Their inactive forms are activated by caspase-1. However, little is known about the mechanism underlying the activation of pro-caspase-1. There is increasing evidence that cathepsin B (CatB), a typical lysosomal cysteine protease, is involved in the pro-caspase-1 activation and the subsequent ma...

Early microglial inhibition preemptively mitigates chronic pain development after experimental spinal cord injury.

Spinal cord injury (SCI) results in the development of chronic pain syndromes that can persist indefinitely and cause reductions in quality of life. Treatment of chronic pain after SCI remains extremely challenging; thus, an important research goal is to determine whether early treatments can attenuate the subsequent development of pain conditions. The current study examined the hypothesis that...

Analgesic effect of α-terpineol on neuropathic pain induced by chronic constriction injury in rat sciatic nerve: Involvement of spinal microglial cells and inflammatory cytokines

Objective(s): Neuropathic pain is a prevalent and debilitating neurological disorder. Ample evidence indicates that microglial cells and inflammatory cytokines are involved in the pathogenesis of neuropathic pain. Alpha-terpineol is a monoterpenoid alcohol with inhibitory effect on inflammatory cytokines. The main purpose of this study was to evaluate the effect of α-t...
